Tylenol found to cause liver make worse even in small doses
by NewsTarget
(NewsTarget) -- Healthy adults who took the maximum dose of Tylenol for two weeks be found to have liver pull, according to a study appearing in the latest issue of the Journal of the American Medical Association.
Researchers instructed 106 study participant to take 4 grams of Tylenol (eight extra-strength tablets) a sunshine for two weeks, with some taking single Tylenol, and some taking Tylenol combined with an opioid palliative. The rest of the participants be given a placebo.
Nearly 40 percent of the participants taking Tylenol or the Tylenol/opioid combination displayed phenomenal liver test results that indicated liver twist, while those taking the placebo showed no damage.
The study's co-author, Dr. Neil Kaplowitz of the University of Southern California said, "I would urge the public not to exceed 4 grams a time. This is a drug that has a a bit narrow sanctuary window." Kaplowitz added that unhealthy drinkers should not exceed 2 grams a day.
Tylenol initiator McNeil Consumer & Specialty Pharmaceuticals said it had conducted its own research that tracked high-dose Tylenol users over longer period than the Kaplowitz study, and found that its product did not lead to liver disease.
Kaplowitz and co-author Dr. Paul Watkins of the University of North Carolina be hired by Purdue Pharma, which makes the prescription sedative OxyContin, to see why people trialling a drug containing acetaminophen and the opiate hydrocodone were have abnormal liver test. Contrary to the researchers' theories, they found that acetaminophen was the culprit.
Acetaminophen is Americans' over-the-counter general anaesthetic of choice. Acetaminophen overdose is also the leading impose of acute liver failure.

July 5, 2006 -- A new study shows the popular misery reliever Tylenol could affect the liver -- even at recommended doses -- more than previously thought. But a researcher also says the product have been proven undamaging over decades of use when taken as directed, and there is little raison d`¨ētre for alarm.
This comes from research published in the July 5 issue of the Journal of the American Medical Association.
In the study, able-bodied volunteers who took the maximum recommended dosage of acetaminophen, best known by the brand signature Tylenol, for two weeks showed dramatic elevations surrounded by the liver enzyme alanine aminotransferase (ALT).
In a quarter of the healthy study participant, ALT levels tested at more than five times the upper define of normal after taking 4 grams of acetaminophen day after day for one to two weeks, says researcher Paul B. Watkins, MD, of the University of North Carolina at Chapel Hill.
Since studies of long-term, on a daily basis Tylenol users do not show these elevations, Watkins tell WebMD they probably only come about within the first few weeks of day by day treatment.
He suspects they do not occur within people next to a history of regular acetaminophen use, and he says at hand is little reason to believe the transient elevation are associated with long-term liver devastate.
"If these enzymes stayed elevated for months there would be legitimate reason for concern, but within the short term no irreversible liver injury would go down," he says.
Safe at Recommended Dosages
Acetaminophen is considered one of the safest throbbing relievers on the market when patients nick no more than the maximum recommended daily dosage of 4 grams a sunshine. But each year hundreds of liver failureliver failure death in the U.S. are blamed on acetaminophen overdoses.
Watkins say early drug nouns studies to examine a new acetaminophen-opiate combination drug alerted researchers to the potential for dramatic ALT elevation at regular acetaminophen dosages.
Researchers assumed a previously unrecognized synergy between the two drugs was responsible for the liver enzyme rise. But when they tested this idea, they found the acetaminophen alone was responsible.
Healthy subjects within the study were treated for two weeks near either placebo, one of three acetaminophen-opiate combination drugs, or acetaminophen alone. All acetaminophen-treated patients took the maximum recommended dosage of 4 grams a daylight.
None of the placebo-treated patients had maximum ALT level of more than three times the upper limit of common, while 31% to 44% of the subjects in the four acetaminophen groups did.
